Triacylglycerol turnover in large and small rat adipocytes: effects of lipolytic stimulation, glucose, and insulin.

چکیده
Rates of lipolysis and reesterification were determined under various conditions in adipocytes from epididymal fat pads of old, spontaneously obese rats and compared to cells from younger, leaner animals. No differences were observed in lipolytic responsiveness to several concentrations (2--50 microM) of the beta-adrenergic agent ritodrine compared to cells from younger, leaner rats. The large cells showed diminished rates of lipolysis, reesterification, and glyceride-glycerol synthesis from glucose of a submaximal but not maximal insulin concentrations, probably reflecting decreased large cell receptor numbers. In both cell types reesterification measured in the presence of ritodrine progressively rose with increasing concentrations of glucose in the medium. At each glucose concentration (0.5--25 mM) rates of reesterification were similar in each cell type. When maximal concentrations of insulin (5 nM) were also added, at low glucose concentrations (less than 5 mM) there was a similar increase in reesterification in large and small cells. No insulin effect in either cell type was observed at high concentrations of glucose. Although fatty acid synthesis from glucose in the large cells was markedly diminished, glyceride-glycerol synthesis was well maintained, correlating well with calculated reesterification rates. In fact, reesterification was found to be quantitatively very important in determining total triacylglycerol turnover in both cell types. High rates of reesterification might not only allow maintenance of triacylglycerol stores, but could also increase metabolic sensitivity to changes in hormonal or substrate concentrations.

Short-term effects of obestatin on hexose uptake and triacylglycerol breakdown in human subcutaneous adipocytes
AIM To study complete dose-dependent effects of obestatin on lipolytic and glucose transport activities in human adipocyte preparations highly responsive to insulin. METHODS Adipocytes were prepared by liberase digestion from subcutaneous abdominal adipose tissue obtained from overweight subjects undergoing plastic surgery. The index of lipolytic activity was the glycerol released in the incu...
متن کاملPartial Inhibition of Adipose Tissue Lipolysis Improves Glucose Metabolism and Insulin Sensitivity Without Alteration of Fat Mass
When energy is needed, white adipose tissue (WAT) provides fatty acids (FAs) for use in peripheral tissues via stimulation of fat cell lipolysis. FAs have been postulated to play a critical role in the development of obesity-induced insulin resistance, a major risk factor for diabetes and cardiovascular disease. However, whether and how chronic inhibition of fat mobilization from WAT modulates ...
متن کاملIsolation of insulin-sensitive phosphatidylinositol-glycan from rat adipocytes. Its impaired breakdown in the streptozotocin-diabetic rat.
In this study an insulin-sensitive glycophospholipid from rat adipocytes was isolated and partially characterized. A material that activated pyruvate dehydrogenase was extracted from rat adipocyte membrane supernatants. Its release was stimulated by insulin and phosphatidylinositol-specific-phospholipase C and its activity was destroyed by nitrous acid deamination. These findings suggested that...
متن کاملPancreastatin inhibits insulin action in rat adipocytes.
Pancreastatin (PST), a regulatory peptide with a general inhibitory effect on secretion, is derived from chromogranin A, a glycoprotein present throughout the neuroendocrine system. We have previously demonstrated the counterregulatory role of PST on insulin action in rat hepatocytes. Here, we are reporting the PST effects on rat adipocytes. PST dose dependently inhibits basal and insulin-stimu...
متن کاملLipolytic effects on diacylglycerol accumulation in human adipose tissue in vitro.
When fragments of rat or human adipose tissue, or isolated adipocytes, are incubated with [14C]glucose in vitro, [14C]diacylglycerol accumulates rapidly: it comprises 20-50% of newly synthesized (14C-labeled) acylglycerols, compared to less than 1% diacylglycerol accumulated in the bulk lipid store in vivo. The experiments reported in this study were performed to test the possibility that agent...
